Light spectra as triggers for sorting improved strains of Tisochrysis lutea

چکیده

It is known that microalgae respond to different light colors, but not at single-cell level. This work aimed assess if colors could be used as triggers sort over-producing cells. Six spectra were used: red + green blue (RGBL), (BL), (RL), (GL), (BRL) and (BGL). Fluorescence-activated cell sorting method was analyse fluorescence BGL RGBL lead the highest fucoxanthin production, while RL showed lowest. Therefore, it hypothesized hyper-producing cells can isolated efficiently under adverse condition (RL). After exposure all for 14 days, top 1% producing sorted. A sorted strain from higher (16–19%) growth rate productivity. study how affected lipid contents productivities. Also, supplied an approach high-fucoxanthin or high-lipid

Continuous selection pressure to improve temperature acclimation of Tisochrysis lutea

Temperature plays a key role in outdoor industrial cultivation of microalgae. Improving the thermal tolerance of microalgae to both daily and seasonal temperature fluctuations can thus contribute to increase their annual productivity. A long term selection experiment was carried out to increase the thermal niche (temperature range for which the growth is possible) of a neutral lipid overproduci...

Proteomic analysis of lipid body from the alkenone‐producing marine haptophyte alga Tisochrysis lutea

Lipid body (LB) is recognized as the cellular carbon and energy storage organelle in many organisms. LBs have been observed in the marine haptophyte alga Tisochrysis lutea that produces special lipids such as long-chain (C37 -C40) ketones (alkenones) with 2-4 trans-type double bonds. In this study, we succeeded in developing a modified method to isolate LB from T. lutea. Purity of isolated LBs ...

Comparative Transcriptome of Wild Type and Selected Strains of the Microalgae Tisochrysis lutea Provides Insights into the Genetic Basis, Lipid Metabolism and the Life Cycle

The applied exploitation of microalgae cultures has to date almost exclusively involved the use of wild type strains, deposited over decades in dedicated culture collections. Concomitantly, the concept of improving algae with selection programs for particular specific purposes is slowly emerging. Studying since a decade an economically and ecologically important haptophyte Tisochrysis lutea (Ti...

miRNA-like duplexes as RNAi triggers with improved specificity

siRNA duplexes, the most common triggers of RNA interference, are first loaded into an Argonaute (Ago) protein and then undergo unwinding via passenger strand cleavage, which requires the slicer activity of the Ago protein. In mammals, only Ago2 out of the four Ago proteins possesses such slicer activity. In contrast, miRNA/miRNA* duplexes often contain central mismatches that prevent slicer-de...
